The hirudin-like factors HLF3 and HLF4-hidden hirudins of European medicinal leeches

Abstract
The hirudin-like factors 3 (HLF3) and 4 (HLF4) belong to a new class of leech-derived factors and are present in specimens of the three European medicinal leeches, Hirudo medicinalis, Hirudo verbana, and Hirudo orientalis, respectively. Here we describe the functional analysis of natural and synthetic variants of HLF3 and HLF4. Whereas the natural variants display only very low or no detectable anti-coagulatory activities, modifications within the N-termini in combination with an exchange of the central globular domain have the potency to greatly enhance the inhibitory effects of respective HLF3 and HLF4 variants on blood coagulation. Our results support previous observations on the crucial importance of all parts (both the N- and C-termini as well as the central globular domains) of hirudin and HLF molecules for thrombin inhibition.

Warum dies für die Hirudotherapie relevant ist
Diese Studie charakterisierte funktionell die hirudin-ähnlichen Faktoren 3 und 4 (HLF3, HLF4) – eine neu beschriebene Klasse von aus Blutegeln gewonnener Faktoren, die in drei europäischen medizinischen Blutegelarten (Hirudo medicinalis, H. verbana und H. orientalis) vorkommen – und fand, dass natürliche Varianten minimale oder nicht nachweisbare antikoagulatorische Aktivität aufwiesen, während gentechnisch veränderte Modifikationen an den N-Termini in Kombination mit einem Austausch der zentralen globulären Domäne die Thrombinhemmung erheblich verstärkten. Diese Arbeit erweitert das Verständnis des Blutegel-Speichelsekretoms (Secretome) unmittelbar, indem sie das bekannte Repertoire an thrombin-zielenden Molekülen über das klassische hirudin hinaus erweitert und aufklärt, wie jeder Strukturbereich – N-Terminus, C-Terminus und zentrale globuläre Domäne – zur inhibitorischen Wirksamkeit beiträgt. Die klinische Relevanz für die Hirudotherapie bleibt indirekt, da die verstärkten antikoagulatorischen Effekte aus gentechnisch veränderten molekularen Modifikationen und nicht aus natürlichen Blutegelsekreten resultierten und das Abstract keine in-vivo- oder therapeutischen Ergebnisdaten liefert.
