Abstract

Platelet function testing is a crucial component in the diagnosis and management of hemostatic disorders in feline patients. Despite its importance, standardized methodologies for assessing platelet function in cats remain underexplored. This study aimed to optimize the conditions for Chrono-log platelet impedance aggregometry (IA) in feline whole blood, focusing on the impact of agonist type (collagen or ADP), agonist concentration, and anticoagulant (citrate or hirudin) on platelet aggregation responses. Whole blood from 10 clinically healthy domestic shorthair cats was prospectively evaluated under varying experimental conditions. Our results indicated that hirudin-anticoagulated samples produced significantly higher platelet aggregation responses compared to those anticoagulated with citrate across all agonist concentrations. A partial dose-dependent relationship was observed with collagen but not with ADP. These findings underscore the importance of selecting appropriate assay conditions for accurate platelet function testing in cats. This study provides insight into the methodology of feline whole blood impedance platelet aggregometry using the Chrono-log analyzer. Malgré son importance, les méthodologies standardisées pour évaluer la fonction plaquettaire des chats demeurent sous-explorées. Cette étude visant à optimiser les conditions d’impédance d’aggrégométrie (IA) plaquettaire du Chrono-log avec du sang entier félin, en mettant l’emphase sur l’impact du type d’agoniste (collagène ou ADP), la concentration d’agoniste et l’anticoagulant (citrate ou hirudine) sur les réponses d’agrégation plaquettaire. Du sang entier de 10 chats domestiques à poil court cliniquement sains a été évalué prospectivement sous diverses conditions expérimentales. Nos résultats indiquent que les échantillons avec l’anticoagulant hirudine produisaient des réponses d’agrégation plaquettaire significativement plus élevées comparativement aux échantillons avec du citrate envers toutes les concentrations d’agonistes. Une relation dose-dépendante partielle a été observée avec le collagène mais pas avec l’ADP. Ces résultats soulignent l’importance de sélectionner des conditions d’analyse appropriées pour des tests précis de la fonction plaquettaire chez les chats.
